Desmin, the intermediate filament protein in striated muscle, is an essential component of the striated muscle cell cytoskeleton. In muscle, the cytoskeleton is a multifunctional, dynamic, and complex structure that serves as a scaffold to maintain the structural integrity and architecture of the cardiomyocyte. However, it serves more than a passive, structural purpose as the cytoskeleton functions as a platform for inter- and intracellular signaling, linking the contractile apparatus so that sarcomeres may function efficiently as a syncytium, and linking those structures in turn to the sarcolemma, cell–cell junctions, the mitochondria, and nucleus. It is not surprising then that mutations in the gene encoding desmin can cause myopathies in general and cardiomyopathy in particular.
